limitation of studies are species differences in metabolism and formation of reactive intermediates<\/li><li>Spontaneous malformation rate is high (3-10%) and variable between different regions, age length of exposure<\/li><li>Time of exposure<\/li><li>Genetic factors (of mother and fetus)<\/li><li>Additive effects of other teratogens<\/li><\/ul><\/li><\/ul>\n\n\n\n<div style=\"height:100px\" aria-hidden=\"true\" class=\"wp-block-spacer\"><\/div>\n\n\n\n<h2 class=\"wp-block-heading\">Teratogens 3<\/h2>\n\n\n\n<ul class=\"wp-block-list\"><li>Teratogenicity is confined to effect of agents on&nbsp;somatic cells&nbsp;(mutagens act on germ cells)<ul><li>However teratogens are often mutagens as well<\/li><\/ul><\/li><li>Majority of birth defects (about 70%) occur with no recognisable causative factor<\/li><li>Recognised teratogenic agents<ul><li>Drug or chemical exposure accounts for about 1% of all fetal malformations (thalidomide, roaccutane, cocaine, alcohol, AEDs)<\/li><li>Infectious agents (rubella, syphilis, cytomegalovirus)<\/li><li>Ionising radiation<\/li><li>Disease (diabetes, lupus, phenylketonuria in miscarriage<\/li><li>cytotoxic drugs and ethanol are exceptions<\/li><\/ul><\/li><li><strong>Organogenesis<\/strong>&nbsp;\u2013 day 17 to day 60, critical period of embryonic development when organs start to develop<ul><li>susceptible to teratogens eg. thalidomide<\/li><\/ul><\/li><li><strong>Histogenesis and functional maturation<\/strong>&nbsp;\u2013 Beyond day 60, functional changes in organs<ul><li>Drugs that interfere with hormones or nutrients inhibit growth or development (not teratogenic)<\/li><li>eg. renal dysfunction by ACE
